The case for and against

Updated Oct 17, 2026

Bull case

Alector is compelling because its ABC blood-brain barrier shuttle could turn the company from a single-asset antibody story into a broader platform for antibody, enzyme, and siRNA delivery across neurodegenerative diseases. The pipeline is strengthened by validated biology and differentiation, including AL137’s 29–32x higher brain exposure in NHPs versus naked antibody, AL050’s Genentech licensing signal, and potent siRNA knockdown data of 58–76% for MAPT and 76–94% for SNCA. Although AL137 still faces clinical execution risk, the 3Q27 healthy-volunteer safety readout offers a clear upcoming catalyst, while the platform’s targeted delivery and cleaner reticulocyte profile could support multiple shots on goal.

Bear case

Alector is still fundamentally exposed to classic clinical-stage biotech risk: its value depends on AL137, AL164, AL062, and AL050 advancing successfully through preclinical and clinical studies and then obtaining regulatory approval in the US and EU. Although the $100M AL050 upfront from Genentech and risk-adjusted milestones and royalties improve liquidity and support the model, the company remains dependent on unproven programs and updated AL137 launch assumptions rather than recurring commercial cash flow. The cited valuation framework also highlights fragility, with only $0.67 of taxed and diluted FY36 GAAP EPS discounted back to FY26 and an NPV that assumes peak revenue achievement, making downside significant if market size, penetration, or pricing disappoints.
