The case for and against

Updated Oct 14, 2026

Bull case

Assembly Biosciences is well capitalized, with the May 22 offering producing $115.0 million gross and about $108.1 million net, plus an expected $75.0 million payment from Gilead in 4Q26, supporting a pro forma runway into 2029. Its outlook is strengthened by external validation and partnership economics in HSV, where GS-1179 was selected to advance and Phase 2 initiation in recurrent genital herpes is expected by the end of 2026, while the optional HIV PrEP setting expands the commercial ceiling beyond suppressive therapy. Fundamentally, the company also has differentiated, oral clinical assets in HDV and cholestatic liver disease, including ABI-6250 and a planned ~140-patient Phase 2 basket study in 1Q 2027, with management citing U.S. peak sales potential of about $2 billion for PBC and $1 billion for PSC.

Bear case

Assembly Biosciences is viewed negatively because its value remains heavily dependent on high-risk clinical and regulatory execution across a narrow set of programs, including ABI-6250, ABI-5366, and ABI-1179, while partner concentration with Gilead amplifies execution risk. Although management reported $226.6M in cash at the end of 1Q 2026 and believes it can fund operations into 2028, the company is not expected to be profitable on a continuing basis until 2032, implying a likely need for additional capital and potential dilution from future financings. In addition, commercialization, reimbursement, manufacturing, and approval-path uncertainty for HBV and herpesvirus assets, plus the fact that ABI-4334 is only a potential partnering upside rather than a core valuation driver, limits confidence in durable fundamental upside.
