Bull case

Black Diamond Therapeutic is well-positioned to succeed in the oncology space due to their deep understanding of cancer genetics, onco-protein structure and function, and medicinal chemistry. Their clinical-stage program, silevertinib, has a covalent MOA with no paradoxical activation, wild-type EGFR sparing, and high brain-penetration which makes it ideal for treating CNS tumors. The company has a potential market of ~4,000 GBM patients in the US, and with sufficient cash to initiate a Phase 2 trial and data expected in 2028, they have the potential to secure an attractive development partnership.

Bear case

Black Diamond Therapeutic is expected to have lower sales for its MasterKey therapy, silevertinib, compared to other similar drugs like osimertinib due to a higher clustering of progression events around 9-11 months. Additionally, with 33% of classical-like patients already seeing success with osimertinib, it is unclear if silevertinib will offer a better alternative. The company's new strategy for treating GBM also faces challenges, as past therapies have failed due to the heterogeneity of EGFR alterations, their paradoxical activation, and poor tolerability, which silevertinib may not address.
