The case for and against

Updated Oct 14, 2026

Bull case

Dianthus Therapeutics is attractive fundamentally because its lead asset class is aimed at validated autoimmune pathways, while the internally developed DNTH312 should improve economics versus the in-licensed DNTH212 and extend franchise IP protection to at least 2047, versus at least 2043 for claseprubart. Management’s strategy also benefits from an experienced clinical team and evidence that DNTH312 produced comparable in vitro potency to claseprubart, plus durable NHP pharmacology with a roughly 22-day half-life and meaningful Ig reductions. With current cash of $1.2B providing runway beyond pivotal claseprubart readouts and key programs such as Phase 2 MMN now tracking to 4Q26, the company appears well positioned to fund multiple shots on goal.

Bear case

Dianthus Therapeutics is viewed negatively because its investment case still depends on a single clinical-stage asset, Claseprubart, while regulatory risk remains elevated and approval may require additional or longer studies if accelerated pathways are not accepted. Although 1Q26 revenue was $0.5M and GAAP EPS of $(0.85) beat consensus, the business is still far from meaningful commercialization, and the company must keep funding development and future launch efforts. Even with $1.2B in cash and a runway into 2030 after the $719M secondary offering, competitive pressure, uncertain labeling, and the need to access capital markets underscore a fragile long-term fundamental outlook.
