The case for and against

Updated Oct 17, 2026

Bull case

Korro Bio is attractive because KRRO-121 targets hyperammonemia with a clear, measurable clinical bar—blood ammonia below 200 μmol/L—making early proof-of-concept easier to assess and potentially broadening use across urea cycle disorders and hepatic encephalopathy regardless of mutation. The first cohort has been dosed in the Phase 1/2 ANCHOR study in Australia, with up to 50 healthy volunteers in Part 1 and up to 15 UCD patients in Part 2, while the AATD program added momentum through >70% SERPINA1 editing and repeat-dosing results above 90% editing. Financially, cash and marketable securities rose to $157.1M at 3/31/26, management guided runway into 2H28, and the net loss improved to $19.6M, supporting continued pipeline execution.

Bear case

Korro Bio is challenged by a valuation that depends on highly speculative assumptions, including a 13% discount rate, 1% terminal growth, and only $225M in estimated EV, while assigning just 20% approval odds to KRRO-121 in UCD and 10% to KRRO-111 in AATD. The outlook remains negative because the thesis is concentrated in a small pipeline, with the analysis explicitly excluding expansion into other programs and warning that KRRO-121 data and clinical progress could disappoint. Long-term dilution risk further weakens the fundamental case, as 20M fully-diluted shares are assumed and any financing needs could pressure per-share value.
