The case for and against

Updated Oct 15, 2026

Bull case

Maze Therapeutics is supported by a compelling genetics-led pipeline that could address large, underdiagnosed markets in CKD and PKU, with APOL1-targeted therapies potentially serving an estimated ~250k U.S. patients with proteinuric kidney disease and ~1 million with high-risk APOL1 genotype. The outlook is further strengthened by MZE829’s preliminary March data showing -49% uACR reductions in AMKD patients without diabetes, a clean safety profile, and a comfortable dose margin at 250mg QD versus higher tolerated doses, while updated HORIZON data in 4Q26 may further validate the asset. MZE782 also adds diversification and a valuation floor, as its Phase 2 PKU study is underway with data expected in 2027 and its convenient oral profile could compete favorably against injection-heavy options like Palynziq.

Bear case

Maze Therapeutics is viewed negatively because its value remains highly contingent on two early clinical assets, MZE829 and MZE782, both of which still face meaningful risks of failure in clinical studies, regulatory approval, and eventual commercialization. Although the company reported encouraging HORIZON Phase 2 data in March 2026, the dataset was small, open-label, and only 12 patients were efficacy evaluable, so the results are not yet strong enough to de-risk the pipeline or prove durable disease modification. Financially, the April registered offering lifted cash to $495M and extended runway into 2029, but the company is still expected to need additional external funding, leaving shareholders exposed to dilution and development delays if capital markets tighten.
