The case for and against

Updated Oct 21, 2026

Bull case

MapLight Therapeutics is viewed positively because ML-007C-MA targets two large central nervous system markets with substantial unmet need, including schizophrenia and Alzheimer’s disease psychosis, where no approved therapies exist for AD psychosis and a differentiated safety profile could support branded pricing despite generic competition. Its clinical profile is encouraging, with favorable safety and tolerability, high target-dose attainment, and potential differentiation from Cobenfy through dosing convenience and reduced peripheral side effects, while the recently completed Phase 2 enrollment and Fast Track designation in Dec 2025 strengthen the development story. The company also has multiple visible catalysts, including 2H 2026 FDA meetings for schizophrenia, a 1H 2027 estimated Phase 3 initiation, and a 2H 2027 VISTA topline readout, while the $150M PIPE financing provides near-term capital support.

Bear case

MapLight Therapeutics is challenged by weak clinical conviction after ZEPHYR missed its primary endpoint, while the data suggest the safety profile in schizophrenia may lose differentiation versus Cobenfy and thereby reduce market penetration. Although management still has roughly $501M of pro forma cash to support programs through 2028, the lowered peak sales assumptions to about $400mn/$2bn in schizophrenia/ADP and a DCF-based price target cut from $43 to $24 underscore deteriorating fundamental expectations. More broadly, the investment case remains highly dependent on future ADEPT and VISTA readouts in difficult neuropsychiatry indications, where small effect sizes, tolerability risk in frail patients, and the need for additional financing or acceptable partnership terms create substantial downside.
