The case for and against

Updated Oct 14, 2026

Bull case

Vaxcyte is viewed favorably because its lead VAX-31 program delivered an unusually strong OPUS-1 readout, meeting all 32 prespecified co-primary endpoints and showing non-inferiority versus PCV20 on 20 of 20 serotypes and versus PCV21 on 17 of 19 at the stringent 0.667 threshold, with a favorable safety profile. This substantially de-risks the adult opportunity in the large $8B+ pneumococcal market, where ACIP expansion to adults >50 years old could broaden demand and VAX-31’s added serotype coverage, including advantages over PCV21, may support competitive positioning. The bull case is further reinforced by VAX-A1’s sizable commercial potential in Group A Strep and by near-term catalysts in 1H27, though the company still faces manufacturing work and a BLA submission not expected until 2028.

Bear case

  • Vaxcyte is facing a negative outlook because its shares are pressured by challenging market conditions, typical financing pressure, and a near-term catalyst path that still leaves roughly eight months before potentially value-inflecting pediatric phase 2 data.
  • The fundamental risk profile is elevated: clinical-stage vaccine development can fail on safety or efficacy, VAX-31 may not differentiate from competing programs, and regulatory outcomes remain uncertain.
  • Commercially, Vaxcyte must displace approved pneumococcal vaccines and build an infrastructure against larger rivals, while the company does not anticipate profitability for several years and may need additional capital.
