The case for and against

Updated Oct 21, 2026

Bull case

Scribe Therapeutics is attractive fundamentally because STX-1150 targets a large, persistent unmet need in ASCVD: poor long-term adherence to chronic LDL-lowering therapy, with more than ~13M secondary prevention patients in the U.S. not at goal and PCSK9-class sales already above $5.1B in 2025. Its preclinical package is compelling, showing >5x greater potency in primary human hepatocytes than NHP hepatocytes, saturated PCSK9 repression at ≥0.6 mg/kg in humanized mice, and >50% LDL-C lowering sustained for two years in NHPs with no significant liver-safety signal. The company also looks financially interesting because its market value is ~$540M versus an estimated ~$400M EV and the Verve acquisition sets a meaningful benchmark for early human PCSK9 proof, while its cash runway into 1H29 supports development through the 1H27 readout.

Bear case

Scribe Therapeutics is fundamentally challenged by the gap between encouraging nonhuman results and unproven human durability, since STX-1150’s repeat dosing in NHPs improved LDL-C lowering from 44% to 57% to 64% but human re-dosing, exposure needs, and long-term safety remain uncertain. The company has no product revenue, expects continued losses, and depends on early clinical readouts expected in 1H27 while management says current capital only funds operations into 1H29, leaving meaningful financing and dilution risk if timelines slip. Commercially, it faces a crowded ASCVD market with established oral and injectable lipid-lowering therapies, limited PCSK9 penetration, and a highly sensitive reimbursement environment that could cap adoption even if efficacy is confirmed.
