The case for and against

Updated Oct 14, 2026

Bull case

Sagimet Biosciences is supported by a differentiated acne franchise built around denifanstat, where 52-week data showed efficacy improving well beyond the Week-12 endpoint and reaching about 58% IGA success with roughly 73% total-lesion and 78% inflammatory-lesion reductions. The fact that efficacy deepened through around Week 24 and remained durable with a clean long-term safety profile strengthens confidence that the drug can be used chronically, potentially expanding patient months on therapy and supporting a better commercial profile than short-course treatments. This also meaningfully de-risks the upcoming AURORA Phase 3 study, while the company’s ~$366M pro forma cash and runway into 2029 provide funding flexibility for denifanstat and TVB-3567.

Bear case

Sagimet Biosciences is viewed negatively because, despite denifanstat’s improved clinical profile in acne, the company still faces meaningful execution risk around replicating efficacy and safety in a broader population, including adolescents, while long-term tolerability and durability remain unproven. Its valuation depends on a DCF using an 11.3% WACC and a 12.5x exit multiple on 2036 estimates, but that framework is undermined by major downside risks from trial failure, regulatory setbacks, competitive pressure, and uncertainties around TVB-3567 and other pipeline assets. Financially, the company is expected to remain loss-making until commercialization and likely through the early launch phase, with a need for additional capital that could dilute shareholders, even as it attempts to penetrate a large U.S. acne market of more than 5 million treatment-seeking patients.
